Abstract

Ionic liquids (ILs) pretreatment is recently becoming an attractive approach for advance the efficiency of enzymatic hydrolysis on lignocellulose. Choline acetate (ChoAc) is not only remarked as biocompatible ILs but also already intensively explored as an effective catalyst to pretreat lignocellulosic biomass. ChoAc could enhance enzymatic saccharification of lignocellulosic biomass according to its capability to deconstruct the compact biocomposite structure of biomass. This working observed hydrolysis activities of cellulase and xylanase which produce extra-cellularly by isolated actinomycetes under the presence of remain IL in pretreated rice straw. The pretreatment was conducted by using the various ratio of ChoAc/biomass (g/g) between 0.0-3.0 and subsequently hydrolysis process was conducting using selected actinomycetes that have the ability to excreted cellulase and xylanase. The ChoAc/biomass ratio was as detected to be 1.5, which achieved the highest activity of cellulase and xylanase at 63.28 u/mL and 79.65 U/mL respectively. The presence of ChoAc over 2.5 g/mL was declining cellulase activities to under 5 U/mL, on the contrary, xylanase activity remains stable at the range 52.23-79.65 U/mL. This investigation indicates the encouraging bio-hydrolysis of biomass in green technology to enhance the yield of the enzymatic saccharification of lignocellulosic biomass in a one-pot technique.
