Abstract

Screening of indigenous bacteria with lipolytic activity from oil-contaminated soil in Bandar Lampung, Indonesia, has been carried out. Identification of bacteria based on morphology, gram staining, and biochemical activity showed that the bacteria were Pseudomonas sp. The exponential phase of bacterial growth on selective media containing olive oil and Tween 80 was formed at 6-20 hours of incubation with the highest lipolytic activity of 60,76 U/ml of enzyme at 18 hours of incubation. This bacteria was methanol tolerant with lipolytic activity of 12,78 U/ml of enzyme at 18 hours incubation and 23,05 U/ml of enzyme at 36 hours of incubation. Enzyme purification using ammonium sulfate precipitation and sephadex G-75 column chromatography showed specific activities of 18.59 U/mg protein and 12.14 U/mg protein, respectively. The lipase produced in this study has temperature kinetics similar to that of bacterial lipases in general, namely the optimum temperature of 450C.
