Orchid Dendrobium sp. is one type of orchid with quite a demand in the orchid market. The drought problem is one of the obstacles to the activity of orchid growth physiologically and morphologically. One way that can be used to develop cultivars of orchid Dendrobium sp., which are resistant to drought stress, is by selecting in vitro plantlet culture with PEG 6000. This study aimed to determine the concentration range of PEG 6000, which was tolerant to drought stress, to analyze the chlorophyll content and stomata index of the orchid plantlet Dendrobium sp. experiencing drought. This research used PEG 6000 with five levels: 0%, 5%, 10%, 15%, and 20%, with five replications. The results showed that the concentration of PEG 6000 was tolerant to drought stress in the orchid plantlet Dendrobium sp., i.e., 20%. The greater the concentration of PEG 6000, the chlorophyll a, b, and total content of the orchid plantlet Dendrobium sp. and the stomata index decreased.
