Abstract

Orchid mycorrhiza is a form of mutualistic association between the roots of orchids with certain fungi that serve as bio-fertilizers. This fungus will infect the orchid through the root which is marked by the presence of hyphal structures in the form of a dense loop within the roots cortex called the pelotons. This study aims (1) to determine whether the four isolates of Orchid mycorrhiza are capable of infecting and performing a symbiosis with the roots of P. amabilis orchid, (2) to determine the best growing medium for acclimatizing P. amabilis orchid plants, (3) to know whether the response of orchid plants toward Orchid mycorrhiza is influenced by the growing medium, and (4) to determine the best growing medium for four types of Orchid mycorhhiza isolates. This research was performed at the Plantation Laboratory, Faculty of Agriculture, Lampung University and the Greenhouse of the Faculty of Agriculture, Lampung University, using a Randomized Group Design (RBD) with factorial treatments (5x2). The first factor is the type of Orchid mycorrhiza (M) isolates, namely control without Orchid mycorrhiza (m0), Orchid mycorrhiza M1 (m1) isolates, Orchid mycorrhiza M9 (m2) isolates, Orchid mycorrhiza M12 (m3) isolates, and Orchid mycorrhiza M14 (m4) isolates. The second factor is the growing medium (T) which consists of two levels, namely cocopeat (t1) and husk charcoal (t2). Each treatment was performed in 4 repetition and each repetition was consisted of 2 orchid seedlings. Orchid seedlings were grouped into 4 based on the number of leaves. The obtained data were tested for various homogeneity by using Bartlett test and additivity by the Tukey test. Data were analysed by using analysis of variance and continued with LSD test at 5% level. The results showed that all tested isolates of M1, M9, M12, and M14 were Orchid mycorrhiza because they were able to infect and performed a symbiosis with P. amabilis orchid roots and form a peloton in the root cortex cells. Cocopeat and husk charcoal growing medium produced the same response to the growth of P. amabilis during acclimatization. The response of orchids toward the administration of Orchid mycorrhiza is not influenced by the growing medium used. Cocopeat and husk charcoal growing medium produced the same response toward the four types of Orchid mycorrhiza isolates.
